The first sign that a dog is in labor is a drop in body temperature.

A dog's normal body temperature is between 101 and 102.5 degrees Fahrenheit. When labor begins, the body temperature will drop to between 99 and 100 degrees Fahrenheit. This drop in temperature is caused by the release of the hormone relaxin, which relaxes the muscles of the cervix and uterus.
